[Biological activity detection of mouse type I soluble interleukin-1 receptor]

Abstract

Objective: To comfirm that the recombinant sIL-1R had biological activity.

Methods: With biotin labed human IL-1 beta as ligand, protein blotting receptor-ligand binding assay and enzyme-linked immunological receptor-ligand binding assay were developed to detect the expression of sIL-1R I gene in insect cells (Sf9). Meanwhile, the blockage assay of IL-1 beta biological activity was used to detect the function of recombinant sIL-1R I.

Results: It showed that the recombinant sIL-1R I possessed immunological activity and ligand binding activity and could be secreted into the medium.

Conclusions: The three assays could be used for detecting biological activity of recombinant sIL-1R I.
